ABSTRACT

Objective:To investigate the effect of miR-129-3p on the proliferation, migration and invasion of hepatocellular carcinoma cells by targeting E2F transcription factor 5 (E2F5).Methods:The expression of miR-129-3p and E2F5 in different liver cancer cell lines and normal liver cell lines were detected by fluorescence quantitative polymerase chain reaction. HepG2 cell lines overexpressing miR-129-3p were constructed, thiazole blue cell proliferation assay and plate cloning test were used to detect the proliferation and clone formation ability of each cell. Cell scratch assay and transwell assay were used to detect cell migration and invasion. Western blotting was used to detect the protein expression. The targeting relationship between miR-129-3p and E2F5 was verified by dual luciferase reporter gene method and western blotting. The experimental groups were as follows: non-transfection (NC) group; transfection control miR-con (miR-con) group; transfection of miR-129-3p (miR-129-3p) group; co-transfection of miR-129-3p and empty vector pcDNA (miR-129-3p+ pcDNA) group; co-transfection of miR-129-3p and pcDNA-E2F5 (miR-129-3p+ pcDNA-E2F5) group.Results:The expression of miR-129-3p in MHCC-97H, HepG2, LM3, Hep3B, PLC, HUH7 were all lower than that of HL-7702, the difference were statistically significant (all P<0.05). The number of cell clones [(86.56±20.84) vs. (511.29±45.03)and(509.78±40.81)], the cell migration rate [(3.03±1.29)% vs. (15.01±2.30)% and(14.99±2.31)%], the relative expression of migration-related protein MMP-2 [(0.51±0.22)vs.(1.87±0.30)and(1.84±0.35)]and the number of transmembrane cells [(33.10±1.58) vs. (101.23±0.31) and (100.96±3.44)] in miR-129-3p group were all decreased when compared with NC group and miR-Con group, the difference was statistically significant ( P<0.05). Dual luciferase reporting assay showed that miR-129-3p can negatively target E2F5 and inhibit its expression. Conclusion:The expression level of miR-129-3p is low in hepatocellular carcinoma cells, overexpression of miR-129-3p can inhibit the malignant biological behavior of hepatocellular carcinoma cells by targeting E2F5.

ABSTRACT

Objective To analyze characteristics and clinical significance of affected and unaffected side plaques in patients with symptomatic middle cerebral artery (MCA) atherosclerosis by three dimensional-sampling perfection with application optimized contrasts using different flip angle evolutions (3D-SPACE) of 3.0 T high-resolution magnetic resonance imaging (MRI).Methods During the period from September 2016 to June 2017,31 patients admitted to the Peking University Aerospace School of Clinical Medicine were enrolled in this study.Routine MRI and high-resolution MRI were performed respectively.Patient's affected and unaffected sides were distinguished based on the conventional MRI on the presence or absence of signs of ischemia.The characteristics of bilateral MCA plaques were studied by high-resolution MRI 3D-SPACE sequences.The stenosis rate,plaque load,eccentricity index were quantitatively analyzed,and the degree of plaque enhancement and remodeling were qualitatively analyzed.Results Thirty-one MCAs in affected and unaffected sides were respectively collected,and 124 MCA plaques and reference plane images were obtained.Among all the patients,tihe MCA in unaffected side was larger in lumen area and had atherosclerotic plaques,and some MCAs had varying degrees of thickening of the wall.The stenosis rate,plaque burden and significantly enhanced plaque proportion of MCA in affected side were significantly higher than those in unaffected side (stenosis rate:76.2% ± 11.5% vs 51.7% ± 13.1%,t =7.801,P<0.01;plaque burden:81.6% ±8.0% vs 56.7% ±13.9%,t =8.614,P<0.01;significantly enhanced plaque proportion:50.0% (13/26) vs 0 (0/26),x2 =17.333,P< 0.01).There was no statistically significant difference in plaque distribution between the two sides.Positive remodeling blood vessels had more vessel area than negative remodeling blood vessels ((13.8 ± 2.9) mm2 vs (11.3 ± 2.8)mm2,t =2.389,P =0.024).Conclusion The high resolution MRI 3D-SPACE sequence clearly shows the MCA wall and plague,and can analyze the enhancement degree and morphological features of affected and unaffected side atherosclerotic plaques simultaneously,thus it is beneficial for risk stratification assessment and individualized treatment for stroke Patients in clinical practice.

ABSTRACT

Objective To investigate the application value of high-resolution magnetic resonance imaging (HR-MRI) as an auxiliary means to evaluate the atherosclerotic stenosis of middle cerebral artery (MCA) before endovascular intervention.Methods Patients with the new infarction or transient ischemic attack in the MCA territories were examined with HR-MRI and digital subtraction angiography (DSA).HR-MRI was used to study the characteristics of atherosclerotic plaques of MCA,qualitatively analyze vascular stenosis rate and plaque load,and qualitatively analyze the plaque location,enhancement degree,and remodeling pattern.The results of HR-MRI examination were used to guide the selection of treatment methods,and the perioperative complications and 3-month outcome of patients treated with intravascular interventional therapy were statistically analyzed.Correlation analysis and consistent evaluation of MCA stenosis rate and stenosis degree measured by DSA and HR-MRI were conducted.Results Atherosclerotic plaques and luminal stenosis in the MCA were found in 25 patients.Among them,13 (52.0%) had positive remodeling,10 (40.0%) had negative remodeling,and 8 (32.0%) had plaques on the sides of perforator arteries.Of the 17 patients who underwent enhanced scan,11 (64.7%) had mild enhancement and 6 (36.3%) had obvious enhancement.Six patients (24%) were treated with endovascular treatment without perioperative complications.Five patients had good outcome and 1 had poor outcome.The stenosis rates of MCA measured by DSA and HR-MRI were 68.05% ± 21.65% and 70.9% ± 20.7%,respectively (t =1.673,P=0.107).The two methods had good correlation (r=0.917,P< 0.001).In addition,in the diagnosis of mild (P =0.500),moderate (P =0.063) and severe (P =0.250) stenoses,DSA and HR-MRI also had good consistency.Conclusion HR-MRI can clearly show the characteristics of the middle cerebral artery lumen and plaque.As a preoperative evaluation means for assisted endovascular treatment of atherosclerotic stenosis,it has an important application value.

ABSTRACT

Objective To monitor intraluminal renal pelvic pressure during retrograde intrarenal surgery ( RIRS ) with syringe irrigation and to investigate the safety of this irrigation method.Methods Seven patients admitted for the flexible ureteroscopic lithoripsy with indwelling nephrostomy tube were enrolled.Two males and 5 femals, with the age from 29 to 58 years (median 48 years).The renal pelvic pressure was measured by the pressure transducer during RIRS.Results In the 7 cases, the mean intra-pelvic pressure( IPP) ranged from 5.1 to 54.8 cm H2 O,the maximum intra-pelvic pressure( IPPmax) ranged from 12 to 158 cmH2O, the time of lithotripsy ranged from 8.25 to 54.73 min (median 23.12 min),the water consumption ranged from 250 to 2300 ml( median water consumption 640 ml) ,and the mean irrigation rate ranged from 27.19 to 40.02 ml/min ( median irrigation rate 31.14 ml/min) , respectively.Conclusions The intraluminal renal pelvic pressure during retrograde intrarenal surgery ( RIRS) with syringe irrigation can be controlled at a low level.The cooperation of pushing water during RIRS by the assistant is crucial.The syringe irrigation method has the advantages of controllable pressure, a rapid adjustment, negative pressure suction in necessity and short duration of high pressure.

ABSTRACT

BACKGROUND:Anatomic medul ary locking (AML) femoral prosthesis is circular cylinder and has satisfactory efficacy. However, some scholars found the complications such as thigh pain, loss of bone at the proximal end of the femur, and wearing-related osteolysis. F2L femoral prosthesis is cone-shaped and also has satisfactory efficacy, but the thigh pain incidence is relatively low. <br> OBJECTIVE:To compare the intermediate-long term results of AML versus F2L in total hip arthroplasty. <br> METHODS:Between November 1997 and January 2005, we retrospectively reviewed 60 patients (66 hips) undergoing total hip arthroplasty using biological femoral prosthesis. At fol ow-up examination, 58 hips in 52 patients were available for clinical and roentgenographic review. 26 AML devices were placed in 24 patients, and 32 F2L devices were placed in 28 patients. The AML group were reviewed with an average of 12.7 years fol ow-up (range 10 years and 3 months to 15 years and 5 months), while the F2L group were reviewed with an average of 9.5 years fol ow-up (range 8 years and 3 months to 11 years and 1 month). The clinical results were evaluated with Harris methods and X-ray examination. Kaplan-Meier analysis was performed to evaluate the survival of femoral component. End point was radiographical loosening or revision of the femoral component for any reason. <br> RESULTS AND CONCLUSION:There were no significant difference between AML and F2L about Harris score in the latest fol ow-up (P>0.05). After surgery, the incidence of thigh pain was significantly lower in F2L group than that in AML group (P<0.05). In AMKL group, the stress-shielding 1 level was observed in 21 hips (81%), and 2 level in five hips (19%);in F2L group, the stress shielding 0 level was observed in 20 hips (62%) and 1 level in 12 hips (38%). There were significant differences between the two groups (P<0.05). The stress shielding showed significant differences between the two groups (P<0.05). The incidence of osteolysis in F2L group was significantly lower than that in AML group (P<0.05). Kaplan-Meier analysis showed that, the survival rate of both AML and F2L components were 1.0 (95%confidence interval:0.98-1.00). Experimental findings indicate that, both AML and F2L femoral prosthesis have a satisfactory long-term efficacy after total hip arthroplasty, and the incidence of thigh pain and osteolysis is significantly lower in F2L group.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the expression of Notch signaling receptors Notchl, Notch3 and its ligand Jaggedl, Jagged2 in tongue squamous cell carcinoma (TSCC).</p><p><b>METHODS</b>mRNA and protein expression levels of tissue samples from 74 cases of tongue cancer patients and human tongue cell line Cal-27 were detected by reverse transcriptase-polymerase chain reaction (RT-PCR), immunohistochemistry and Western blot. Its relationship with cell proliferation and clinical pathology was analyzed.</p><p><b>RESULTS</b>mRNA and protein expression were detected in tongue cancer tissues, adjacent tissues and cell lines. Notchl and Notch3 protein expression in tongue cancer was higher than the adjacent tissues. Jaggedl and Jagged2 protein expression in tongue cancer and adjacent tissues had no difference. Notchl and Notch3 protein had correlation with tongue cancer clinical staging. Pathway protein expression had no correlation with pathological grade, age, gender. Notchl protein expression in lymph node metastasis-positive cases was higher than in lymph node metastasis-negative cases. The expression of Notch3 and Jagged2 had correlation. Jaggedl expression grade in metastasis-positive cases was higher than in negative cases.</p><p><b>CONCLUSION</b>Notch signaling molecules have active expression in TSCC and may play important roles in tongue cancer development.</p>

ABSTRACT

Uncoupling protein 4 (UCP4) is a member of the multigenic uncoupling proteins (UCPs), specific expressing in the cerebral cortex and hippocampus. UCP4 plays an important role in Parkinson's disease, multiple sclerosis, epilepsy, stroke, brain trauma and other central nervous system diseases by uncoupling, decreasing mitochondrial membrane potential,regulating Ca2+ homeostasis and oxidative stress. This article reviews UCP4 and its roles in the central nervous system diseases in order to provide certain basis for the development of UCP4targeted medication.

ABSTRACT

Objective To evaluate the clinical efficacy of pingyangmycin (PYM) injection on infantile hemangioma located in the parotid gland region. Methods Twelve patients were treated by intralesional injection of PYM. When necessary, the injections were repeated at an interval of one week, but not more than 3-4 sessions within a therapeutic period. Normally, the secondary therapeutic period was performed 1 month later. The general and local adverse responses were recorded and the clinical outcomes were assessed with a follow-up of 1 to 3 years. Results Complete clinical resolutions were achieved in 10 patients. 2 patients received one injection, 3 patients received 2 injections, 3 patients received 3 injections, and 2 patients received 4 injections. The remaining 2 patients with partial resolution received 6 and 7 injections respectively. No clinical recurrence was observed during the follow-up of 1 to 3 years. No ulcerations or postoperative sears in injection regions were presented. The function of facial nerve was remained normality in all patients. The systematic side effects included transient pyrexia and poor appetite appeared in partial patients. No allergy cases were found. Conclusion Treatment of infantile hemangioma located in parotid gland region with PYM injection reveals a high rate of complete clinical resolution, with fair cosmetic results and short treatment time, and it does not damage the facial nerve or form local scar.The treatment time of PYM injection seems to be positively related to size of the lesions.

ABSTRACT

Objeetive To investigate the outcome of microvascular reconstruction of the tongue with anterolateral thigh flaps in the treatment of middle-late stage tongue cancer patients. Methods From December 2003 to March 2007,nine patients underwent simultaneous reconstruction of the tongue and oral floor defects with anterolateral thigh flaps after resection of squamous cell carcinoma of tongue.The flaps ranged from 7 cm×10 cm to 10 cm×12 cm in size,and were adjusted to the defect of the tongue and oral floor.The vascular pedicle included descending branch of the lateral femoral circumflex artery and the accompanying veins.The outcome of reconstruction was evaluated by follow-up examinations,considering the contour and mobility of the reconstructed tongues,the swallowing function and the speech function.Results All of the donor sites were closed directly,with minimal donor-site morbidity. All patients recovered unevenffully from surgery,with no immediate postoperative complications:no flap necrosis,no wound infection or wound dehiscence.The transplanted flaps survived well.The average follow-up period was 18 months.During the follow-up period there was no tumor recurrence and the contour of the reconstructed tongues showed sufficient bulk.The patients demonstrated good mobility of the reconstructed tongue.The swallowing and speech function recovered satisfactory.Two months postoperatively the patients were able to ingest a solid or semisolid diet,and six months postoperatively the patients developed intelligibe language.Conclusion The anterolateral thigh flaps are suitable and reliable for the microsurgical reconstruction of the large defects caused by middle-late stage tongue cancer.

ABSTRACT

Adrenomedullin (AM) is a novel vasoactive peptide. The actions of AM include vasodilatation, reduction of arterial pressure, inhibition of vascular smooth muscle cell transference and proliferation, diuretic and natriuretic, inhibition of aldsterone secretion. Furthermore AM modulates extra cellular matrix deposition. So adrenomedullin plays an important role in the regulation of cardiovascular remodeling. This paper reviews the relationship between AM and cardiovascular remodeling.
